FITC-Labeled Human Siglec-2, His Tag (Cat. No. SI2-HF2H6) is expressed from human HEK293 cells. It contains AA Asp 20 - Arg 687 (Accession # P20273-1). It is the FITC labeled form of Human Siglec-2, His Tag (Cat. No. CD2-H52H8).

蛋白結(jié)構(gòu)（Molecular Characterization）

This protein carries a polyhistidine tag at the C-terminus.

The protein has a calculated MW of 77.0 kDa. The protein migrates as 100-120 kDa under reducing (R) condition (SDS-PAGE) due to glycosylation.

標(biāo)記（Labeling）

The primary amines in the side chains of lysine residues and the N-terminus of the protein are conjugated with FITC using standard chemical labeling method. The residual FITC is removed by molecular sieve treatment during purification process.

背景（Background）

B細(xì)胞受體CD22也被稱為唾液酸結(jié)合Ig樣凝集素2（Siglec-2）、B淋巴細(xì)胞粘附分子（BL-CAM）、T細(xì)胞表面抗原Leu-14，屬于免疫球蛋白超家族和Siglec（唾液酸結(jié)合Ig-like凝集素）家族。CD22介導(dǎo)B細(xì)胞與B細(xì)胞的相互作用，并可能參與B細(xì)胞在淋巴組織中的定位。Siglec-2/CD22結(jié)合唾液酸化糖蛋白，其中之一是CD45。Siglec2/CD22通過與Src家族酪氨酸激酶的相互作用在正向調(diào)節(jié)中發(fā)揮作用，也可能通過其SH2結(jié)構(gòu)域招募細(xì)胞質(zhì)磷酸酶來(lái)充當(dāng)抑制性受體，SH2結(jié)構(gòu)區(qū)通過信號(hào)分子的去磷酸化阻斷信號(hào)轉(zhuǎn)導(dǎo)。
