GDNF分子背景

膠質(zhì)細(xì)胞源性神經(jīng)營養(yǎng)因子（GDNF）是一種神經(jīng)營養(yǎng)因子，可增強(qiáng)多巴胺能神經(jīng)元的存活和形態(tài)分化，并增加其高親和力多巴胺攝取。GDNF是一種糖基化、二硫鍵合的同二聚體，是轉(zhuǎn)化生長因子β超家族的遠(yuǎn)親。GDNF家族成員（由GDNF、neurturin、artemin和persephin組成）在結(jié)構(gòu)上與轉(zhuǎn)化生長因子β相似。與通過受體絲氨酸-蘇氨酸激酶發(fā)出信號的TGF-β超家族的其他成員不同，GDNF家族配體通過受體酪氨酸激酶Ret激活細(xì)胞內(nèi)信號級聯(lián)。GDNF家族配位體結(jié)合GDNF家族受體α（GFRalpha），然后GDNF家族-配體-GFR-alpha復(fù)合物結(jié)合并刺激Ret的自磷酸化。
