CD20分子背景
B淋巴細(xì)胞抗原CD20也稱為B淋巴細(xì)胞表面抗原B1、白細(xì)胞表面抗原Leu-16、跨膜4結(jié)構(gòu)域亞家族A成員1和MS4A1，是一種活化的糖基化磷蛋白，從B前相（CD45R+、CD117+）開始在所有B細(xì)胞表面表達(dá)，濃度逐漸增加，直到成熟。CD20在B細(xì)胞發(fā)育的所有階段都有表達(dá)，除了第一個(gè)和最后一個(gè)階段；它從晚期pro-B細(xì)胞通過記憶細(xì)胞存在，但不存在于早期pro-B細(xì)胞或漿母細(xì)胞和漿細(xì)胞上。在B細(xì)胞淋巴瘤、毛細(xì)胞白血病、B細(xì)胞慢性淋巴細(xì)胞白血病和黑色素瘤癌癥干細(xì)胞中發(fā)現(xiàn)。該蛋白沒有已知的天然配體，其功能是實(shí)現(xiàn)最佳的B細(xì)胞免疫反應(yīng)，特別是針對(duì)T非依賴性抗原。懷疑它在細(xì)胞膜中充當(dāng)鈣通道。CD20/MS4A1是單克隆抗體（mAb）利妥昔單抗、伊維單抗和托西莫單抗的靶點(diǎn)，這些抗體都是治療所有B細(xì)胞淋巴瘤和白血病的活性藥物。CD20/MS4A1的缺陷是免疫缺陷共同變量5型（CVID5）的原因；也稱為由于CD20缺陷引起的抗體缺乏癥。CVID5是一種原發(fā)性免疫缺陷，其特征是抗體缺乏、低丙種球蛋白血癥、反復(fù)細(xì)菌感染和無法對(duì)抗原產(chǎn)生抗體反應(yīng)。
