This novel anticancer agent (FW = 403.55 g/mol), also known as (2E,4E)- 5-((1S,2S,4aR,6R,7S,8S,8aS)-7-hydroxy-2,6,8-trimethyl-1,2,4a,5,6,7,8,8aoctahydronaphthalen- 1-yl)-2-methyl-N-(pyridin-3-ylmethyl)penta-2,4- dienamide), is a powerful cell growth inhibitor (GI50 = 12 nM) and equally potent Golgi disruptor (EC50 = 27 nM), inhibiting activation of ADPribosylation factors (or Arfs) by its guanine nucleotide exchange factors (GEFs). Arfs are Ras superfamily small GTPases that play a major role in mediating vesicular transport in the secretory and endocytic pathways. Molecular dynamics (MD) simulations suggest that AMF-26 binds to the contact surface of the Arf1-Sec7 domain near the site for brefeldin A. AMF-26 strongly affected membrane traffic, including the cis-Golgi and trans-Golgi networks, and the endosomal systems. When tested with a panel of 39 cancer cell lines, the GI50 was consistently below 40 nM. The total synthesis of AMF-26 has also been described.
