1-Boc-吡唑-4-硼酸頻哪醇酯屬于吡唑類衍生物,主要用作藥物分子和生物活性分子的合成中間體。在合成轉(zhuǎn)化中,吡唑環(huán)上的硼單元是一個非常實用的官能團轉(zhuǎn)化前體,硼單元可以很容易地轉(zhuǎn)化為氮原子,也可以很容易被氧化成羥基,或者通過Suzuki偶聯(lián)反應(yīng)在硼單元的位置連接上一個芳基或者烷基基團。合成方法
將氯(1,5-環(huán)辛二烯)(1,1'-菲羅啉)銥四氫呋喃加合物(5.8 mg, 0.01 mmol, 0.5 mol%)和聯(lián)硼酸頻哪醇酯加入到一個敞口的反應(yīng)瓶中,用一個帶隔膜的螺旋蓋密封小瓶,通過在真空下進行三次循環(huán)抽真空來對小瓶進行氮氣置換。在第二個燒瓶中制備頻那醇硼烷(0.050 mL,44.1 mg,0.344 mmol)在四氫呋喃(50 mL,0.0067 M)中的儲備溶液。將儲備溶液(1.4毫升,1當(dāng)量的頻那醇硼烷)注射到氯(1,5-環(huán)辛二烯)(1,1'-菲羅啉)銥四氫呋喃加合物和硼酸頻哪醇酯的混合體系中去,將所得的反應(yīng)混合物加熱到65度反應(yīng)1小時,直到形成暗紅色溶液。將0.336克1H-吡唑-1-甲酸叔丁酯直接加入暗紅色溶液中,讓其反應(yīng)若干個小時,通過GC、GC-MS和TLC監(jiān)測反應(yīng)的進展。當(dāng)反應(yīng)完成后,冷卻小瓶,通過硅藻土過濾除去固體沉淀,得到的濾液真空下蒸發(fā)旋干,用二氯甲烷溶解所得的殘余物,通過柱色譜法(0:100-1:1 乙酸乙酯和正己烷)提純分離殘余物即可得到目標(biāo)產(chǎn)物1-Boc-吡唑-4-硼酸頻哪醇酯。
