Step 2: A mixture of 2-amino-3-(methoxycarbonyl)pyrazine1-oxide (5.50 g, 32.5 mmol) and POCl3 (15.2 mL,163 mmol) in DMF (30 mL) was heated at100°C for16 h. After cooling to room temperature, the mixture was diluted with H2O (300 mL) and adjust to pH = 7 with solid NaHCO3. The resulting mixture was extracted with ethyl acetate (2 x 200 mL). The combined organic layers were washed with brine (100 mL), dried and concentrated to dryness. The residue was purified by column chromatography (silica gel,100-200 mesh,10% ethyl acetate in petroleum ether) to obtain methyl 3-amino-5-chloropyrazine-2-carboxylate (940 mg,15.4% yield).
