1 Example 1Preparation of isochroman (Compound 2)
A solution of paraformaldehyde (16.0g, 506mmol) was added 2-phenylethyl alcohol (1) (41.5g, 334mmol) and stirred, then added dropwise concentrated hydrochloric acid 36-38% (of 37 mL), warmed to 36 . 20h reaction, the reaction is complete the basic raw material, the end of the reaction, was allowed to stand under cooling to room temperature. The organic layer was separated by a separating funnel. 150mL of water was added to the aqueous layer extracted with dichloromethane (2 × 150mL), the organic layers combined with the organic phase was washed successively with 5% sodium hydrogen carbonate (80mL), water (2 × 150mL) and saturated brine (150mL) . The organic phase was dried over anhydrous sodium sulfate overnight, filtered, and the filtrate was concentrated under reduced pressure to give a pale green oil 45.3g, 99% yield.
