Steps:
General Method I of Synthesis (Taking Compound DXL-A-10 as an example)
To a 100 mL round-bottom flask with 25 mL of isopropanol, m-iodotoluene (2.18 g, 0.01 mol) was added, then anhydrous piperazine (1.76 g, 0.02 mol), cuprous iodide (0.5 g, 25 mmol), potassium phosphate (4.66 g, 17.5 mmol) and ethylene glycol (1.5 mL) were added in sequence, and reacted under reflux and argon atmosphere for 18 hours, until TLC (PE: EA = 20; 1, DCM: MeOH = 10: 1) detection showed the reaction was completed. After filtration, the solvent was removed under reduced pressure, 20 mL of water was added, and extraction was carried out with 30 mL of chloroform for three times. The extracts were combined, washed with saturated brine and water, dried over anhydrous sodium sulfate, concentrated to dryness and subjected to column chromatography (EA: MeOH: NH3·H2O = 25: 3: 2) to obtain 0.5 g of a colorless oily product 2-22, yield: 23%.1H NMR (400 MHz, CDCl3): δ 7.18 (t, J = 7.7 Hz, 1H), 6.74 (dd, J = 17.8, 8.7 Hz, 3H), 3.14 (s, 8H), 2.35 (s, 3H).
