Scheme 1 Charge NMR tubes (17 x 0.3 cm i.d., ~1.7 mL volume) with 0.20 g of Au and seal with a high-vacuum Teflon stopcock. Load the NMR tube with a 0.60 mL CD3CN solution containing nBuNC (10.0 mM), amine or alcohol (30.0 mM), and R3NO (35.0 mM) in a glovebox.Heat the mixture at 60°C in an oil bath.Monitor the reaction progress by 1H NMR or take aliquots (~50 μL) periodically by a syringe and analyze by GC.Sublime the amine N-oxides under reduced pressure to reduce the chance of introducing any water into the reaction.Handle in a glovebox and bring nBuNC into the box in a nitrogen-packed ampule to obtain the product. The synthesis route is shown in Fig. 1[1].

Scheme 2 Charge a 2 L flask with 4 mol of tolylene diisocyanate. Add 1 mol of the alkyl amine to the mixture under stirring. Heat the mixture to the boiling point. Distill off the resulting product until the amount reaches what is theoretically possible. Pour the remaining hot mixture quickly into a large metal container (strong foaming with subsequent solidification is possible). Purify the product additionally by vacuum distillation. The synthesis route is shown in Fig. 2[2].
Chemical Properties of Butyl Isocyanate
Butyl isocyanate, with the chemical formula C4H9NCO, stands out for its unique molecular structure, characterized by the isocyanate group (-N=C=O) bonded to a butyl group. This structural feature imparts reactivity that is central to its wide range of applications. The compound is a colorless liquid at room temperature, with a distinctive pungent odor that is a hallmark of isocyanates.
Physically, butyl isocyanate has a boiling point of approximately 154-156°C and a density slightly less than water. It is sparingly soluble in water but exhibits good solubility in organic solvents, making it versatile in various chemical processes. Its reactivity with compounds containing active hydrogen atoms, such as water, alcohols, and amines, enables it to form polymers, elastomers, and other valuable materials.
Safety is paramount when handling butyl isocyanate due to its toxic and potentially irritating effects on the skin and respiratory system.
