RFect小核酸轉(zhuǎn)染試劑是知名科學(xué)家崔坤元博士領(lǐng)導(dǎo)我公司研發(fā)團(tuán)隊(duì)在美國西雅圖實(shí)驗(yàn)室研發(fā)成功的一種新型的小核酸轉(zhuǎn)染試劑。RFect可用來轉(zhuǎn)染siRNA、miRNA mimics、miRNA inhibitor 等200bp以內(nèi)的小分子RNA,轉(zhuǎn)染細(xì)胞包括絕大多數(shù)貼壁生長的細(xì)胞,如一般細(xì)胞株、腫瘤細(xì)胞株等。目前,無論國外還是國內(nèi),轉(zhuǎn)染試劑的主要成分均為脂質(zhì)體或聚乙烯亞胺(Polyethylenimine, PEI),這兩種成分都具有很大的細(xì)胞毒性,并且轉(zhuǎn)染效果不好。RFect采用新型的動(dòng)物源性的納米材料,毒性很低并且擁有非常高的轉(zhuǎn)染性能。與其它品牌的小核酸轉(zhuǎn)染試劑相比,RFect的細(xì)胞轉(zhuǎn)染陽性率一般在90%以上,Lamin A/C 基因抑制效率在95%以上,而其它品牌轉(zhuǎn)染試劑的細(xì)胞轉(zhuǎn)染陽性率一般不超過70%,Lamin A/C 基因抑制效率不超過75%。RFect細(xì)胞毒性很低,轉(zhuǎn)染細(xì)胞死亡率不到10%,而其它品牌試劑的轉(zhuǎn)染細(xì)胞死亡率一般在30%以上。血清對轉(zhuǎn)染效果沒有影響,不必刻意添加或更換培養(yǎng)液。本產(chǎn)品適用于細(xì)胞株轉(zhuǎn)染,原代細(xì)胞轉(zhuǎn)染,請選擇RFectPM小核酸轉(zhuǎn)染試劑。
產(chǎn)品特點(diǎn)
·細(xì)胞轉(zhuǎn)染性能:細(xì)胞轉(zhuǎn)染陽性率高達(dá)90%以上,基因敲除效果明顯,A549細(xì)胞Lamin A/C 基因敲除效率在95%以上;
· 極低的細(xì)胞毒性:轉(zhuǎn)染細(xì)胞死亡率不到10%,大大降低了因細(xì)胞毒性對實(shí)驗(yàn)結(jié)果的影響;
· 轉(zhuǎn)染細(xì)胞范圍廣,絕大多數(shù)貼壁細(xì)胞株都能獲得比較理想的轉(zhuǎn)染結(jié)果。

Different cell lines were transfected with scrambled or Lamin A/C siRNA (final concentration: 10nM) using RFect In this and all the following figures, all the cells are harvested 48 hours after transfection and Lamin A/C knockdown was measured by qRT-PCR.

A549 cell lines were transfected with Lamin A/C siRNA (final concentration: 10nM) using RFect siRNA transfection reagent. RFect concentrations added to the cells exceeding transfection concentration of 20 times, the cells still do not produce significant toxicity, which makes these product suitable to optimization. Lamin A/C knockdown was measured by qRT-PCR.
