TRAILR2分子背景

腫瘤壞死因子受體超家族成員10B（TNFRSF10B）也稱為TNF相關(guān)凋亡誘導(dǎo)配體受體2（TRAILR2）、死亡受體5（DR5）、CD262、KILLER，是TNF受體超家族的成員，含有細(xì)胞內(nèi)死亡結(jié)構(gòu)域。TNFRSF10B/DR-5在成人和胎兒組織中廣泛表達(dá)；在腫瘤細(xì)胞系中高度表達(dá)。TRAILR2/CD262/DR5是細(xì)胞毒性配體TNFSF10/TRAIL的受體。TRAIL-R2/TNFRSF10B的銜接分子FADD（一種含有死亡結(jié)構(gòu)域的銜接蛋白）將胱天蛋白酶-8募集到活化的受體上。由此產(chǎn)生的死亡誘導(dǎo)信號復(fù)合物（DISC）進(jìn)行胱天蛋白酶-8蛋白水解激活，啟動隨后介導(dǎo)凋亡的胱天蛋白酶（天冬氨酸特異性半胱氨酸蛋白酶）級聯(lián)反應(yīng)。CD262/DR5促進(jìn)NF-kappa-B的激活。DR5對ER應(yīng)激誘導(dǎo)的細(xì)胞凋亡至關(guān)重要，并受p53/TP53的調(diào)節(jié)。
