DKK1分子背景

dickkopf相關(guān)蛋白家族（DKK-1、-2、-3和-4）的成員是分泌蛋白，具有兩個(gè)富含半胱氨酸的結(jié)構(gòu)域，由接頭區(qū)隔開。DKK1通過抑制WNT信號(hào)通路參與胚胎發(fā)育，以高親和力與LRP6結(jié)合，并阻止Frizzled-WNT-LRP6復(fù)合物對(duì)Wnts的反應(yīng)形成。DKK1與細(xì)胞表面受體Kremen形成三元復(fù)合物時(shí)，促進(jìn)LRP6內(nèi)化和降解。DKK1在發(fā)育過程中不僅起頭部誘導(dǎo)劑的作用，還調(diào)節(jié)關(guān)節(jié)重塑和骨形成，這表明DKK1在類風(fēng)濕性關(guān)節(jié)炎和多發(fā)性骨髓瘤的發(fā)病機(jī)制中起著重要作用。最近的研究報(bào)告稱，DKK1從一個(gè)確定的發(fā)育時(shí)間點(diǎn)開始影響眼睛發(fā)育，并且通過β-catenin介導(dǎo)的Pdgfrα和E-cadherin表達(dá)對(duì)晶狀體與表面外胚層的分離至關(guān)重要。
