BK-1361 (cycli (RLsKDK)) is a cyclic peptide composed of six amino acids, which contains a D-type amino acid. It acts as an effective selective inhibitor of ADAM8, with an IC50 of 120 nM; BK-1361 (cycle (RLsKDK)) did not show significant activity against ADAM 9,10,12. BK-1361 (cycli (RLsKDK)) inhibits CD23 shedding in cell-based shedding assay, IC50 is 182 nM, reduces migration/invasion of pancreatic cancer cells, reduces ERK1/2 and MMP activation, and causes changes in cell morphology; A mouse model of PDAC that simultaneously reduces tumor burden and metastasis of implanted pancreatic tumor cells, and provides clinical symptom and survival improvement indicators driven by Kras (G12D).
