Raltegravir是一種有效的integrase (IN)抑制劑，作用于WT和S217Q PFV IN，在無(wú)細(xì)胞試驗(yàn)中IC50分別為90 nM和40 nM。它對(duì)HIV-1 IN的選擇性比對(duì)其他相關(guān)Mg2+依賴性酶要高1000倍以上，如HCV聚合酶、HIV反轉(zhuǎn)錄酶、HIV RNaseH、人類α-, β-, γ-聚合酶。

特性 Raltegravir 是第一個(gè)被批準(zhǔn)的人類免疫缺陷病毒1型（HIV-1）整合酶抑制劑。

體外研究活性 PFV整合攜帶S217H替換的IN，之后對(duì) Raltegravir 的敏感性降低10倍，IC50為900 nM。PFV IN 具有10% WT 活性，且被Raltegravir抑制，IC50為200 nM,說(shuō)明與 WT IN相比，對(duì)整合酶（IN）鏈轉(zhuǎn)移抑制劑(INSTI)的敏感性降低約2倍。S217Q PFV IN 對(duì) Raltegravir的敏感性至少與WT 酶相似。[1] Raltegravir由葡萄糖醛酸化，而不是肝臟代謝而來(lái)。Raltegravir 在體外作用于人類T 淋巴細(xì)胞培養(yǎng)物，有效作用于HIV-1，抑制達(dá)95%時(shí)濃度為31±20 nM。Raltegravir作用于 CEMx174細(xì)胞，也有效作用于HIV-2,IC95為6 nM。Raltegravir 代謝主要通過(guò)葡萄糖醛酸化。葡萄糖醛酸化酶的強(qiáng)誘導(dǎo)劑UGT1A1顯著降低 Raltegravir 濃度。Raltegravir 微弱抑制肝細(xì)胞色素 P450。Raltegravir不會(huì)誘導(dǎo) CYP3A4 RNA 表達(dá)或 CYP3A4依賴的睪丸激素 6-β-羥化酶活性。[2]在鎂和鈣存在時(shí)，Raltegravir細(xì)胞擴(kuò)散性降低。[3]Raltegravir 和相關(guān)的HIV-1整合酶 (IN) 鏈轉(zhuǎn)移抑制劑 (INSTIs) 高效阻斷病毒復(fù)制。[4] Raltegravir 作用于急性感染人類淋巴 CD4+ T細(xì)胞系MT-4和 CEMx174, 高效抑制SIVmac251復(fù)制，EC90處于低納摩爾范圍。[5]

為了定量分析鏈轉(zhuǎn)移，供體DNA底物通過(guò)退火 HPLC 級(jí)寡核苷酸 5′-GACTCACTATAGGGCACGCGTCAAAATTCCATGACA 和 5′-ATTGTCATG GAATTTTGACGCGTGCCCTATAGTGAGTC而形成。反應(yīng)(40 μL) 含0.75 μM PFV IN, 0.75 μM 供體 DNA, 4 nM (300 ng) 超螺旋 pGEM9-Zf(?) 靶 DNA, 125 mM NaCl, 5 mM MgSO4, 4 μM ZnCl2, 10 mM DTT, 0.8% (vol/vol) DMSO, 和 25 mM BisTris 丙烷–HCl, pH 7.45。加入指定濃度Raltegravir。加入2 μL PFV IN （在150 mM NaCl, 2 mM DTT, 和 10 mM Tris-HCl, pH 7.4中稀釋）開(kāi)始反應(yīng),在 37oC下反應(yīng)1小時(shí)后，加入25 mM EDTA 和 0.5% (wt/vol) SDS終止反應(yīng)。反應(yīng)產(chǎn)物通過(guò)與 20 μg 蛋白酶 K 在 37oC下消化30分鐘而脫蛋白，隨后使用乙醇 沉淀,在 在1.5％瓊脂糖凝膠上分離，然后通過(guò)用溴化乙錠染色觀察。通過(guò)實(shí)時(shí)定量PCR, 使用 Platinum SYBR Green qPCR SuperMix 和三種引物 : 5′-CTACTTACTCTAGCTTCCCGGCAAC, 5′-TTCGCCAGTTAATAGTTTGCGCAAC, 和 5′-GACTCACTATAGGGCACGCGT定量分析整合產(chǎn)物。PCR 反應(yīng) (20 μL) 含0.5 μM 每種引物和1 μL 稀釋的整合反應(yīng)產(chǎn)物。在95oC下進(jìn)行5分鐘變性，在CFX96 PCR儀上進(jìn)行35次循環(huán)，再在95oC下變性10秒，在56oC下退火30秒，在68oC下延伸1分鐘。在INSTI存在時(shí)，使用連續(xù)稀釋的WT PFV IN 反應(yīng)，獲得標(biāo)準(zhǔn)曲線。
