產(chǎn)品描述 Bleomycin Sulfate硫酸博來霉素是一種糖肽類抗生素,一種DNA 合成抑制劑,也是一種鱗狀細(xì)胞癌(SCC)抗癌劑,在UT-SCC-19A細(xì)胞中IC50為4 nM。Bleomycin sulfate是一種DNA 合成抑制劑。
體外研究(In Vitro)
體外研究活性

UT-SCC-12A和 UT-SCC-12B均對(duì)Bleomycin sulfate耐藥,IC50分別為14.2 nM 和13 nM。[1]肺泡巨噬細(xì)胞與0.01 μg/mL到1μg /mL Bleomycin sulfate培養(yǎng)18小時(shí)顯著比巨噬細(xì)胞單獨(dú)培養(yǎng)分泌更多的成纖維細(xì)胞生長(zhǎng)因子。Bleomycin sulfate刺激的巨噬細(xì)胞持續(xù)產(chǎn)生大量成纖維細(xì)胞生長(zhǎng)因子,即使在移去Bleomycin sulfate,用新鮮培養(yǎng)基(不含Bleomycin sulfate)取代之后,該作用依然存在。Bleomycin sulfate刺激肺泡巨噬細(xì)胞導(dǎo)致的纖維素生長(zhǎng)因子分泌被環(huán)己酰亞胺,和5-脂氧合酶抑制劑NDGA(去甲二氫愈創(chuàng)木酸)和BW755c抑制,表明其不僅需要蛋白質(zhì)合成,也需要花生四烯酸代謝的5-脂氧合酶途徑活性完全表達(dá)。[2] Bleomycin sulfate (400 µg/mL)培養(yǎng)24小時(shí),降低NTera-2細(xì)胞的活性,增加caspase-3,-8和-9的活性,增加Bax和細(xì)胞質(zhì)細(xì)胞色素C水平,并降低Bcl-2水平。[3]對(duì)于不穩(wěn)定性畸變,Bleomycin sulfate對(duì)ADIPO-P2細(xì)胞的致染色體斷裂作用在處理后至少持續(xù)10天。此外,暴露于Bleomycin sulfate的細(xì)胞處理10天后,端粒融合體的外觀表明Bleomycin sulfate能夠誘導(dǎo)延遲的端粒失穩(wěn)。[4]

方法

ADIPO-P2細(xì)胞在D-MEM高葡萄糖培養(yǎng)基中生長(zhǎng),用20%胎牛血清,青霉素(100 U/mL) 和鏈霉素 (100 μg/mL)增補(bǔ),在37 °C 和5% CO2大氣環(huán)境中培養(yǎng)。細(xì)胞在包含1.5×105 cells/mL的TC25 Corning培養(yǎng)瓶中單層培養(yǎng)。對(duì)于每個(gè)實(shí)驗(yàn),建立兩個(gè)培養(yǎng)瓶,一個(gè)用于對(duì)照組,一個(gè)用于處理培養(yǎng)。在對(duì)數(shù)生長(zhǎng)期,ADIPO-P2細(xì)胞用2.5 μg/mL Bleomycin sulfate以30分鐘脈沖處理。建立平行對(duì)照組,不暴露于Bleomycin sulfate。暴露時(shí)間和Bleomycin sulfate濃度根據(jù)先前實(shí)驗(yàn)室中進(jìn)行的Bleomycin sulfate處理哺乳動(dòng)物細(xì)胞的研究進(jìn)行選擇。Bleomycin sulfate脈沖處理結(jié)束時(shí),細(xì)胞用Hank's平衡鹽溶液清洗兩次,并保存在新鮮培養(yǎng)基中直到采集。處理后5個(gè)傳代培養(yǎng)期間,將細(xì)胞持續(xù)保持在培養(yǎng)基中。當(dāng)培養(yǎng)基中融合(培養(yǎng)基中大約4×105 cells/mL)時(shí),進(jìn)行傳代培養(yǎng)。為估計(jì)細(xì)胞生長(zhǎng),傳代培養(yǎng)時(shí),細(xì)胞通過胰蛋白酶化收集,每等份大約200 μL,用0.4%臺(tái)盼藍(lán)著色,測(cè)定活細(xì)胞數(shù)量。隨后細(xì)胞懸浮在新鮮培養(yǎng)基中,重新分配到包含1×105 cells/mL的新培養(yǎng)瓶中繼續(xù)生長(zhǎng)。將剩余的細(xì)胞丟棄或置于另外的培養(yǎng)瓶中進(jìn)行細(xì)胞遺傳學(xué)分析,這在處理結(jié)束后18小時(shí)或第10天進(jìn)行。為分析染色體突變,培養(yǎng)的最后3小時(shí)將colchicine (0.1 μg/mL)加入到細(xì)胞培養(yǎng)基。染色體的制備根據(jù)標(biāo)準(zhǔn)程序進(jìn)行。采集后,將細(xì)胞低滲震動(dòng),固定在甲醇:乙酸(3:1)中,涂布在載玻片上,進(jìn)行PNA-FISH。進(jìn)行兩個(gè)獨(dú)立的實(shí)驗(yàn)。
