體外研究活性 | 5 μM Obatoclax作用于SK-Mel5細胞完全抑制Mcl-1參與的Bak修復(fù),且阻斷由Mcl-1引起的抗ABT-373誘導(dǎo)的KB/Bcl-2細胞凋亡。 [1] Obatoclax是擬BH3,可以與許多Bcl-2家族成員結(jié)合,包括Bcl-2, Bcl-xL,和Mcl-1。Obatoclax特異取代Mcl-1的激活BH3域,隨后引起B(yǎng)ak低聚反應(yīng)和細胞色素C釋放調(diào)節(jié)的細胞凋亡。Obatoclax可有效作用于缺乏Bcl-xL或Bcl-xL低表達的細胞系,Obatoclax作用于所有強表達Mcl-1, Bcl-2,和Bcl-xL的細胞系時顯示出較低的細胞毒性。Obatoclax抑制多發(fā)性骨髓瘤細胞(MM)系如KMS12PE, KMS18, MY5等等,IC50值為52到1100nM。當(dāng)有濃度為150 nmol的抗細胞毒素劑IL-6 或IGF-1存在時,也可觀察到抑制作用。 [2] Obatoclax加強腫瘤壞死因子相關(guān)凋亡誘導(dǎo)配體(TRAIL)調(diào)節(jié)的細胞凋亡。 [3] |
