To a stirred solution of N(alpha)-Cbz-L-asparagine (10 g, 37.5 mmol) dissolved in DMF (50 ml) was added DCC (7.8 g, 37.5 mmol) and N-hydroxysuccinimide (4.4 g, 37.5 mmol) and the mixture was heated at 80 C for 6 h. DMF was evaporated under reduced pressure and the residue was dissolved in ethyl acetate (100 ml) and filtered to remove DCU. The filterate was washed with water (2 x 100 ml), brine (100 ml) and dried (Na2SO4). The product obtained after evaporation of the solvent was purified by silica gel column chromatography using 50 % ethyl acetate in petroleum ether to give 4.2 g of the product as white solid, mp 66-69 C; IR (KBr) 3395, 3177, 2936, 2749, 1723, 1689, 1519, 1260, 1177 cm-1; 1H NMR (300 MHz, CDC13) 5 2.81-2.89 (m, 1 H), 3.04-3.13 (m, 1 H), 4.33-4.40 (m, 1 H), 5.11 (s, 2 H), 4.63 (brs, 1 H), 7.34 (s, 5 H), 8.54 (brs, 1 H). |
