N-(9-Fluorenylmethoxycarbonyl)-4-bromo-D-phenylalanine (5, 96 mg, 0.21 mmol) was added and the mixturewas kept stirring. PdCl2 (10 mol %, 4 mg) was added and nitrogen was bubbledthrough the reaction mixture for 15 min. Na2CO3 (66 g, 0.62 mmol) was thenadded into the reaction mixture whereupon the flask was filled with nitrogenand heated at 66 C for 3 h. When the TLC showed completion of the reactionwater (20 mL) was added into the reaction mixture which was then extractedwith ethyl acetate (3 30 mL). The combined organic layers were washed withbrine-water (1:1, 2 25 mL), dried over Na2SO4 and concentrated to drynessunder reduced pressure. The crude product was purified by columnchromatography (2-3% MeOH in DCM containing 0.1% AcOH) to affordcompound 6 (87 mg, 81%). |
