Trimethoprim exerts its antibacterial effect by selectively inhibiting the enzyme dihydrofolate reductase in bacteria. By blocking this enzyme, Trimethoprim disrupts the folate metabolism in bacteria, leading to a deficiency of essential precursors for DNA and RNA synthesis. It is particularly useful in treating urinary tract infections caused by organisms such as Escherichia coli, Proteus mirabilis, and Klebsiella pneumoniae. Additionally, it is employed in the treatment of respiratory tract infections, including bronchitis and pneumonia, when caused by susceptible bacteria. Skin and soft tissue infections, as well as some gastrointestinal infections, can also be treated with Trimethoprim, either alone or in combination with other antibiotics.
