Thiamine mononitrate is a stable nitrate salt form of thiamine (vitamin B1). It occurs as a white crystalline powder and is prepared from thiamine hydrochloride. It is used as a food or nutritional supplement and is the preferred form of the vitamin for food fortification. Thiamin mononitrate is used in the pharmaceutical industry to treat beriberi and general malnutrition or malabsorption. Thiamine can be found naturally in foods like grains, yeast, molasses, pork and animal organ meats. Dairy, eggs and legumes have smaller amounts.
Though thiamine is found naturally in foods, thiamine mononitrate is not. Thiamine mononitrate is synthesized by removing a chloride ion from thiamin hydrochloride and mixing the final product with nitric acid. Thiamine hydrochloride is hygroscopic (water-absorbing) whereas the mononitrate has almost no hygroscopic properties. For this reason, the mononitrate is the more stable form of the vitamin in fortified flours and cereals.

Production method

Vitamin B1 is naturally found in rice bran, bran, lean meat, peanuts and other foods, and the pure product is usually prepared by chemical synthesis. Excessive acetamidine hydrochloride is condensed with α-dimethoxy-β-methoxy-propanitrile in alkaline (methanol and sodium methanol) media to 3, 6-dimethyl-1, 2-dihydro-2,4,5, 7-tetrazonium naphthalene, and then hydrolyzed at 98-100℃, and then opened under alkaline conditions to form 2-methyl-4-amino-5-aminomethacil; Then it reacts with carbon disulfide and ammonia water, then condenses with acetic acid-γ-chloro-γ-levulyl ester, then hydrolyzes in hydrochloric acid at 75-78℃ and synthesizes cyclothiamine hydrochloride, and finally neutralized with ammonia water, oxidized with hydrogen peroxide and acidified with hydrochloric acid to obtain thiamine hydrochloride.

Vitamin B1 is an important water-soluble vitamin to maintain the normal function of the nervous system and digestive system. It participates in the formation of coenzymes in the body and maintains sugar metabolism.

Thiamine mononitrate (CAS NO.532-43-4) is applicable to vitamin B1 deficiency, maintain normal metabolism and nerve conduction function. It is also used in adjuvant therapy for indigestion, neuropathy, etc.
