Selenocysteine is an amino acid found in a few enzymes, such as glutathione peroxidase, thyroxine 5' -deiodinase, thioredoxin reductase, formate dehydrogenase, glycine reductase, and some hydrogenases. The structure of selenocysteine is similar to that of cysteine, except that the sulfur atom is replaced by selenium. Proteins that contain selenocysteine residues are called selenoproteins. In the genetic code, the code for selenocysteine is UGA (opal stop codon), which is commonly used as a stop codon. However, if there is a SElenoCysteine Insertion Sequence (SECIS) in the mRNA, UGA is used as the code for selenocysteine. SECIS sequence is determined by a secondary structure formed by a specific nucleotide sequence paired with a base. In true bacteria, SECIS comes directly after the UGA codon, in the same reading frame as UGA. In archaea and eukaryotes, SECIS directs multiple UGA codons encoding selenocysteine residues in the 3' -untranslated region of mRNA (3'-UTR). When cell growth is deficient in selenium, the translation of selenium proteins is halted at the UGA codon, becoming an incomplete and non-functional protein. Like other amino acids in cells, selenocysteine has a specific tRNA. This tRNASec differs from other standard trnas in a few ways, most notably having an eight-base (bacterial) or nine-base (eukaryotic) receiving stem, a long variable arm, and several highly conserved base replacements. tRNASec is initially loaded with a serine by the serine-TRNA ligase, but this Ser-tRNASec is not used for translation because it is not recognized by the usual translation factors (EF-Tu in bacteria, eEF-1α in eukaryotes). This seryl can be replaced by a selenocysteine synthetase containing pyridoxal phosphate to selenocysteyl. Finally, this Sec-tRNASec specifically binds to another translation extension factor, SelB, or mSelB, and is delivered to the ribosome that is translating selenoprotein mRNA.
