PluriSln-1 (91396-88-2) is an inhibitor of stearoyl-coA desaturase (SCD1). This stearoyl-CoA sesaturase 1 inhibitor, or SCD1 (FW = 213.24 g/mol;
CAS 91396-88-2; Solubility: 43 mg/mL DMSO; <1 mg/mL H2O), also
known as NSC 14613 an 4-pyridinecarboxylic acid, 2-phenylhydrazide,
selectively eliminates human Pluripotent Stem Cells (hPSCs), while sparing
a large array of progenitor and differentiated cells, thus providing a
promising approach for minimizing the tumorigenic risk from residual
undifferentiated cells. Closer examination identified PluriSIn #1 as an
SCD1, thereby indicating a role for lipid metabolism in hPSC survival.
PluriSIn #1 is likewise cytotoxic to mouse blastocysts, again suggesting a
dependence on oleate in cells in a pluripotent state. SCD is key and highly
regulated enzyme which catalyzes the D9-cis desaturation of a range of
palmitoyl- and stearoyl-CoA to palmitoleoyl- and oleoyl-CoA respectively.
